چکیده

Article Information Brinjal is an important vegetable crop grown worldwide. A field survey conducted during 2012 in Kerala state of South India, the little leaf disease affected brinjal plant was observed in one of the home gardens located at Nemmara (Palakkad district). The diseased and healthy samples were collected; genomic DNA was isolated and 16SrRNA gene amplified by performing nested PCR using universal primers pairs P1/P7 and R16F2n/R2 respectively. The association of Phytoplasma was detected in all the three symptomatic brinjal samples and in Phytoplasma affected Periwinkle used as positive control. An expected amplicon size of 1250bp from nested PCR was purified and sequenced. The nucleotide sequence analysis (nBLAST) of 1250bp fragment of sequence KL-50(KP027530) revealed 97% similarity with Candidatus phytoplasma trifolii (JX104336). Phylogenetic analysis and study of virtual RFLP gel pattern of the sequence also revealed the association of Candidatus phytoplasma trifolii.

A Rapid Protocol of Crude RNA/DNA Extraction for RT-qPCR Detection and Quantification of 'Candidatus Phytoplasma prunorum'

Many efforts have been made to develop a rapid and sensitive method for phytoplasma and virus detection. Taking our cue from previous works, different rapid sample preparation methods have been tested and applied to Candidatus Phytoplasma prunorum ('Ca. P. prunorum') detection by RT-qPCR. A duplex RT-qPCR has been optimized using the crude sap as a template to simultaneously amplify a fragment ...

Detection of ‘Candidatus Phytoplasma brasiliense’ in a new geographic region and existence of two genetically distinct dnaK genotypes

In September 2007, a peach tree (Prunus persica) displaying yellowing symptoms indicative of phytoplasma infection was sampled in Quba region of Azerbaijan. A phytoplasma was detected in the diseased peach tree by nested PCR amplification of its 16Sr DNA with universal primers for phytoplasmas. Phylogenetic analyses of the amplified 16S rDNA showed that the phytoplasma infecting peach tree corr...

'Candidatus Phytoplasma mali', 'Candidatus Phytoplasma pyri' and 'Candidatus Phytoplasma prunorum', the causal agents of apple proliferation, pear decline and European stone fruit yellows, respectively.

Apple proliferation (AP), pear decline (PD) and European stone fruit yellows (ESFY) are among the most economically important plant diseases that are caused by phytoplasmas. Phylogenetic analyses revealed that the 16S rDNA sequences of strains of each of these pathogens were identical or nearly identical. Differences between the three phytoplasmas ranged from 1.0 to 1.5% of nucleotide positions...

"Candidatus phytoplasma australiense," a new phytoplasma taxon associated with Australian grapevine yellows.

A phytoplasma was detected in naturally diseased 'Chardonnay' grapevines exhibiting symptoms of Australian grapevine yellows disease. The use of PCR designed to amplify phytoplasma DNA resulted in detection of phytoplasma DNA in all of the diseased plants examined; no phytoplasma DNA was detected in healthy seedling grapevines. The collective restriction fragment length polymorphism (RFLP) patt...
